Patients have been waiting for this advance, even if they didn’t know it.” That has been Asymmetrex® President and CEO, James L. Sherley, M.D., Ph.D.’s, continuing refrain, even before the company first reported kinetic stem cell (KSC) counting in 2020. KSC counting is Asymmetrex®’s computational simulation method for routinely quantifying tissue stem cells based on how they produce mature cells in cell cultures.

Since its 2020 report, Asymmetrex® has worked on making KSC counting more accessible for general use in stem cell research, stem cell medicine, cell and tissue biomanufacturing, and pharmaceutical drug development. Because of their central role in organ and tissue maintenance, stem cells are important factors in the development of diseases, such as cancer, and in aging. In addition to improving the effectiveness of stem cell treatments for human patients, quantifying stem cells can improve medical treatments for animals, as well as reduce the use of animals for human drug evaluations.

Using its KSC counting method, Asymmetrex® scientists discovered the existence of mathematical algorithms that can be used to calculate the number of stem cells in a tissue cell preparation by taking a sample of the preparation and culturing it for 72 hours. This simple and relatively inexpensive cell culture procedure is widespread in research and testing laboratories. So, with access to Asymmetrex®’s stem cell counting algorithms, many laboratories can begin quantifying their stem cells as a routine practice.

To advance this development in stem cell science and medicine, this month, Asymmetrex® made five stem cell-counting algorithms available as free rapid stem cell-counting calculators on its website. The calculators provide simple instructions for use. When users enter generic cell counting data from the prescribed 72 hours of general cell culture, the calculators instantly return the corresponding stem cell fraction of the sample. The first set of calculators provided can be used for commonly isolated preparations of human blood stem cells and human fat-derived stem cells. Hundreds of calculators exist for different tissues and different culture conditions.

Asymmetrex® is making the new calculators freely available to support its longer-term vision of using the algorithms to power the first laboratory instruments for quantifying therapeutic stem cells. The future wide deployment of such stem cell counters in tissue cell research labs, stem cell transplant centers, cord blood stem cell banks, cell and tissue biomanufacturing facilities, and pharmaceutical drug evaluation labs will remove a major impediment to the current rate of progress and success for each of these groups, which is that their current operations are blind to the number or dosage of a critical element in many of their processes, the stem cells.
